# Pittsburgh Pelvic Floor Research Program

## Abstract

The purpose of this application is to request supplementary funds to complete ongoing commitments to the
Pelvic Floor Disorders Network (PFDN) at the University of Pittsburgh in 2021-2022. Our site has a
longstanding track record of successful contribution to multicenter studies of urinary and fecal incontinence,
and pelvic organ prolapse. The University of Pittsburgh is particularly well suited to be a clinical site in the
PFDN because of our volume, research infrastructure and track record, basic and translational experience and
expertise. Magee-Womens Hospital (MWH) is the hub of the Women’s Health Service Line for the 40 hospital
University of Pittsburgh Health System, serving a very large aging population of women. Our site brings to the
PFDN expertise in urogynecology, physical therapy, urology, gastroenterology, geriatrics, and biomechanical
engineering. We offer the PFDN unique technical expertise in development of finite element models of the
vagina and its supportive tissues after surgery, biochemical and biomechanical impact of meshes on the
vagina, dynamic 3D MRI and transperineal ultrasound of the pelvic floor, and tissue regenerative techniques.
This expertise is key to completion of the aims of the 2 ongoing translational projects of the PFDN: Defining
Mechanisms of Anterior Vaginal Descent (DEMAND) trial, and the Biofabrication of vaginal support using
vaginally derived cells: a Feasibility study (BioLIFT). In this application for supplementary funds to
UGHD069006 we described our plan for completion of the aforementioned translational projects, and our
research personnel staffing and management plan for recruitment and retention into the Mixed Urinary
Incontinence: Mid-urethral Sling vs. Botox A (MUSA) trial (NCT04171531), SUPeR (NCT01802281) and
ASPIRe (NCT02676973) clinical trials.
