# Precision Interception of Aggressive Prostate Cancer in African American Men

## Abstract

Summary 
The Biospecimen and Biomarker Core (BBC) provides key functions for biosample collection, processing, 
evaluation, storage, and biomarker generation to all three P20 projects and the DRPs. The BBC will provide 
multiple pathology services, including detailed histopathologic review of tissues; characterization of normal and 
tumor tissue; characterizing inflammatory infiltrate; identifying early lesions and prostatic intraepithelial 
neoplasia (PIN); annotating well-characterized frozen and paraffin tissue samples in the tissue bank; and 
providing sections and samples of tissues for project investigators. The BBC will provide state of the art 
imaging services and quantitation of nucleic acids and proteins. The BBC will generate a clinically-linked 
archival paraffin tissue series of human prostate cancer, including construction of a tissue microarrays (TMA), 
immunohistochemistry (IHC), tissue coring for nucleic acid analysis, and image analysis of biomarkers. We will 
further provide a variety of services critical to successful molecular analysis of mouse and human prostatic 
tumors including: histopathologic review and quality control analysis of all tumor samples utilized in 
experimental studies; slide macrodissection of paraffin-embedded tissues to ensure high neoplastic cellularity 
for samples utilized in experimental studies; laser capture microdissection (LCM) to provide ultra-pure tumor 
samples and microscopic sub-populations of cells (such as immune cell populations); optimization of 
techniques in nucleic acid extraction tailored to the experimental goals of the P20 investigators; performance 
and analysis of routine and highly multiplexed IHC and immunofluorescence (IF) stains on TMAs of human 
prostate adenocarcinomas; optimization and validation of antibodies to known and novel proteins for use in 
human IHC; performance and analysis of a broad range of IHC stains in human tissues in direct support of the 
projects. The BBC will also provide resources and technologies for molecular characterization that are required 
for the projects’ research activities. The BBC will adapt and implement new resources and technologies to 
meet the scientific goals of the projects. We provide platforms for high throughput IHC and IF with 
sophisticated image analysis instrumentation and tools. We will also provide genomics services including the 
use of targeted cancer panels (e.g., Oncopanel), RNASeq, Nanostring, Affymetrix arrays, and other related 
tools as required by the projects. Finally, the BBC will manage tissue banking, molecular output, and clinical 
data for translational research. The BBC will manage the central data repository for tissue and biosample 
banking, as well as integration with the clinical data resource for the projects. The BBC is essential to the
