Deciphering Wnt-Ror signaling in cytoskeletal regulation and tissue shape control

Abstract

Supplement Summary I am requesting an administrative supplement for the purchase of a BD Accuri C6 Plus Flow Cytometer. This cytometer is equipped with two lasers and an automatic sampler capable of handling 24 samples at once. This equipment will be crucial for completing the goals of my NIGMS R35 grant (R35GM119574) by enhancing the throughput of a novel reporter assay that our lab developed, under the auspices of this R35 grant, to investigate the biochemical mechanisms of the Wnt5a-Ror morphogenetic pathway. By coupling this reporter assays with a newly optimized gene-replacement paradigm, we have begun to analyze a large number of mutant proteins associated with dysfunction of the pathway and Robinow syndrome pathogenesis. Our workflow has now exceeded the capacities of the currently available flow cytometers which can no longer handle the volume of analyses efficiently, hence the new need for an automated system. The Department of Cell Biology and Human Anatomy at UC Davis School of Medicine is committed to provide space to house the cytometer and funds for instrument service. In summary, the requested flow cytometer will greatly accelerate the NIGMS-funded projects in my lab, as well as those of other NIH investigators in the department and the greater UC Davis community.
