Administrative supplement to purchase a MerMade 4 oligonucleotide synthesizer for the large-scale production of modified RNA substrates

Abstract

ABSTRACT: Our NIH R35 ESI MIRA entitled ‘Selectivity and regulation of mRNA demethylation by iron- dependent dioxygenases’ focuses on understanding the structural and molecular determinants of AlkBH-family RNA demethylase enzymes that play key roles in RNA function and some human cancers. A critical challenge to carrying out some of the proposed structural experiments has been in obtaining sufficient quantities of modified RNA oligonucleotide substrates for crystallographic screening and biophysical binding assays. This administrative supplement requests funds to purchase an LGC MerMade 4 oligonucleotide synthesizer that will transform and greatly accelerate our ability to access key modified RNA oligonucleotide reagents to support multiple goals of our MIRA projects.
